PF-06939999 is an orally available small molecule inhibitor of protein arginine N-methyltransferase 5 (PRMT5), developed by Pfizer, with potential antiproliferative and antineoplastic activities. It inhibits the methyltransferase activity of PRMT5, leading to decreased levels of monomethylated and dimethylated arginine residues in histones H2A, H3, and H4. This modulation affects gene expression involved in cell proliferation, potentially reducing the growth of rapidly proliferating cancer cells[6][8]. Docetaxel is a taxane-class chemotherapeutic agent that promotes microtubule assembly from tubulin dimers and stabilizes microtubules by preventing depolymerization. This disrupts normal mitotic functions essential for cell division, resulting in inhibition of cancer cell growth[1][3]. The combination is being investigated primarily for non-small cell lung cancer (NSCLC) but may also be relevant to other solid tumors[5].
