PF-07062119 + anti-PD-1 + anti-VEGF

PF-07062119 + anti-PD-1 + anti-VEGF is a combination regimen under investigation for advanced or metastatic gastrointestinal cancers, including colorectal, gastric, and esophageal adenocarcinomas. The combination consists of PF-07062119 (a bispecific T cell engager targeting guanylyl cyclase C, GUCY2C, and CD3), an anti-PD-1 monoclonal antibody (specifically PF-06801591, sasanlimab), and an anti-VEGF monoclonal antibody (bevacizumab). PF-07062119 redirects T cells to GUCY2C-expressing tumor cells to induce cytotoxicity, anti-PD-1 blocks the PD-1/PD-L1 interaction to enhance anti-tumor T cell response, and anti-VEGF inhibits VEGF-mediated angiogenesis to limit tumor vascular supply. This three-drug regimen was evaluated together in a Phase 1 clinical trial for safety, tolerability, pharmacokinetics, and preliminary anti-tumor activity in patients with GI cancers, but development was terminated for strategic reasons.
