Overview

PF-07062119 + anti-VEGF is a combination therapy formerly under development by Pfizer for the treatment of advanced or metastatic gastrointestinal cancers, including colorectal, gastric, and esophageal adenocarcinomas. PF-07062119 is a bispecific T-cell engager (BiTE) designed to bind Guanylyl Cyclase C (GUCY2C) on tumor cells and CD3 on T cells, facilitating T-cell mediated cytotoxicity. The addition of an anti-vascular endothelial growth factor (anti-VEGF) agent, specifically bevacizumab in clinical trials, aims to enhance antitumor activity through antiangiogenic mechanisms and potential modulation of the tumor microenvironment. Clinical evaluation occurred in a Phase 1 study (NCT04171141), but Pfizer discontinued the development of PF-07062119 in early 2024.
