PF-07104091 + atirmociclib is a novel combination therapy being developed by Pfizer for HR+/HER2- breast cancer. This combination pairs PF-07104091, a selective CDK2 inhibitor, with atirmociclib (PF-07220060), a first-in-class selective CDK4 inhibitor. The combination is designed to maximize inhibition of both kinases while sparing CDK6 to potentially reduce hematologic adverse events like neutropenia. This approach aims to counter cyclin E-mediated resistance to CDK4/6 inhibitors and provide greater efficacy than either inhibitor alone. The combination has shown synergistic efficacy across HR+/HER2- breast cancer models, including in palbociclib-resistant tumors, with tumor regression rather than tumor stasis.
