This combination pairs PF-07104091, a selective CDK2 inhibitor, with atirmociclib (PF-07220060), a first-in-class selective CDK4 inhibitor. This approach aims to counter cyclin E-mediated resistance to CDK4/6 inhibitors and provide greater efficacy than either inhibitor alone. The combination has shown synergistic efficacy across HR+/HER2- breast cancer models, including in palbociclib-resistant tumors, with tumor regression rather than tumor stasis.
