PF-08046049 + SGN-BB228 is an investigational bispecific antibody therapy designed for the treatment of advanced melanoma and other difficult-to-treat or metastatic solid tumors. The drug conditionally delivers 4-1BB costimulation to T cells specifically in the presence of CD228 (melanotransferrin)-expressing tumor cells, aiming to enhance anti-tumor immune activity while minimizing peripheral toxicity. Mechanistically, it functions as a first-in-class CD228-directed costimulatory Antibody Anticalin bispecific that provides potent, CD228-conditional 4-1BB (tumor necrosis factor receptor superfamily member 9) costimulation to tumor-infiltrating T cells, driving CD8+ T cell effector differentiation and contributing to tumor killing. The therapy is being evaluated in a Phase 1 clinical trial for patients with advanced melanoma and several other solid tumor types.
