PF-08046054 + pembrolizumab is an investigational combination therapy for advanced solid tumors. PF-08046054 (also known as SGN-PDL1V) is a PD-L1-directed antibody-drug conjugate (ADC) that consists of a monoclonal antibody targeting PD-L1, covalently linked via a protease-cleavable maleimidocaproyl-valine-citrulline (mc-vc) linker to the microtubule-disrupting agent monomethyl auristatin E (MMAE). The ADC enables selective delivery of MMAE to PD-L1-expressing tumor cells; the intended mechanisms are cytotoxicity, immunogenic cell death, and bystander effect, while PD-1/PD-L1 checkpoint inhibition is considered unlikely. Pembrolizumab is a monoclonal antibody checkpoint inhibitor targeting PD-1. The combination aims to enhance antitumor immune responses and direct cytotoxicity in metastatic or unresectable solid tumors[1][2][3][4][6].
