Overview

PF-08634404 + chemotherapy is an investigational combination therapy being developed by Pfizer for the treatment of advanced gastroesophageal cancers, including gastric, gastroesophageal junction, and esophageal adenocarcinomas. The regimen consists of PF-08634404, a bispecific monoclonal antibody, and standard-of-care chemotherapy. PF-08634404 simultaneously targets the Programmed Cell Death Protein 1 (PD-1) immune checkpoint and the Vascular Endothelial Growth Factor (VEGF) pathway. By inhibiting PD-1, the drug enhances T-cell mediated anti-tumor immune responses, while VEGF inhibition suppresses tumor-associated angiogenesis and may improve the delivery of chemotherapy by normalizing tumor vasculature. This combination is currently being evaluated in Phase 3 clinical trials as a first-line treatment for several solid tumors.
