Overview

PF-3512676 + gemcitabine + cisplatin is a **combination regimen** investigated for the treatment of advanced non-small-cell lung cancer (NSCLC). PF-3512676 (also known as CpG 7909 or ProMune) is a synthetic oligodeoxynucleotide that acts as a Toll-like receptor 9 (TLR9) agonist, activating plasmacytoid dendritic cells and B lymphocytes, thereby stimulating innate and adaptive immune responses to enhance antitumor immunity. Gemcitabine is a nucleoside analog used as a cytotoxic chemotherapeutic agent, and cisplatin is a platinum-based chemotherapeutic agent that causes DNA crosslinking and cell death. The rationale for this combination was to augment the presentation of tumor antigens generated by chemotherapy and enhance immune response via PF-3512676. Phase III trials demonstrated that adding PF-3512676 to gemcitabine/cisplatin did not improve clinical efficacy and increased toxicity; as a result, development of this combination was discontinued for NSCLC[1][4][5].
