Overview

PF-3512676 + paclitaxel + carboplatin is a combination therapy investigated for the treatment of advanced non-small cell lung cancer (NSCLC). PF-3512676 (formerly known as CPG 7909) is a synthetic oligodeoxynucleotide that acts as an agonist of Toll-like receptor 9 (TLR9), stimulating innate immune responses and potentially enhancing antitumor immunity. Paclitaxel is a microtubule-stabilizing agent that inhibits cell division, while carboplatin is a platinum-based chemotherapeutic that causes DNA crosslinking and apoptosis in rapidly dividing cells. The rationale for combining these agents was to augment the efficacy of standard chemotherapy with immunostimulatory effects from PF-3512676. However, phase III clinical trials showed no improvement in overall survival or progression-free survival with the addition of PF-3512676 to paclitaxel/carboplatin compared to chemotherapy alone, and increased toxicity was observed[2][4][5].
