Overview

PfAMA1-FVO[25-545] is a recombinant protein malaria vaccine candidate developed by the African Malaria Network Trust (AMANET). The vaccine consists of the ectodomain (amino acids 25-545) of the apical membrane antigen 1 (AMA-1) derived from the *Plasmodium falciparum* FVO clone. Produced in the yeast *Pichia pastoris*, the recombinant protein is adsorbed to Alhydrogel (aluminum hydroxide) as an adjuvant. AMA-1 is a critical protein located on the surface of merozoites that facilitates the invasion of host erythrocytes. By eliciting an immune response against this antigen, the vaccine aims to generate antibodies that block parasite entry into red blood cells, thereby preventing or mitigating malaria infection. It has been evaluated in Phase 1 clinical trials for safety and immunogenicity in malaria-endemic regions, specifically in healthy adults in Mali.
