The Pfizer-BioNTech COVID-19 Vaccine, Bivalent (also known as Comirnaty Bivalent) is an mRNA-based vaccine developed to provide broad protection against COVID-19. It contains two messenger RNA (mRNA) components: tozinameran, which encodes the spike protein of the original SARS-CoV-2 strain, and famtozinameran, which encodes the spike protein of the Omicron BA.4 and BA.5 variants. The mRNA is nucleoside-modified and encapsulated in lipid nanoparticles (LNPs) to facilitate delivery into host cells. Upon intramuscular injection, the mRNA is translated by host cells into spike proteins, which then trigger an adaptive immune response, including the production of neutralizing antibodies and the activation of T-cells. This bivalent formulation was specifically designed to address waning immunity and the increased transmissibility of emerging Omicron subvariants.
