Pfs230D1M-EPA + AS01 is a transmission-blocking vaccine candidate against Plasmodium falciparum malaria. It consists of recombinant Pfs230 domain 1 (Pfs230D1M), a subdomain of a surface antigen found on gametocytes, gametes, and zygotes in the mosquito stage of Plasmodium falciparum. This antigen is conjugated to a recombinant Pseudomonas aeruginosa ExoProtein A (EPA) and adjuvanted with AS01, a liposome-based adjuvant system containing the immune-enhancers MPL (3-O-desacyl-4'-monophosphoryl lipid A) and QS21 (a saponin molecule purified from the bark extract of Quillaja saponaria Molina tree). The vaccine is designed to interrupt malaria transmission by inducing antibodies that target the sexual stages of the parasite. When a mosquito takes a blood meal from a vaccinated individual, these antibodies prevent the parasite from developing in the mosquito, thereby blocking transmission to other humans.
