Overview

PGDM1400 + PGT121 is an investigational combination of two broadly neutralizing monoclonal antibodies (bNAbs) developed for the prevention and treatment of HIV-1 infection. Both are recombinant human IgG1 monoclonal antibodies that target distinct sites on the HIV-1 envelope glycoprotein trimer. PGDM1400 targets the V2 apex region, while PGT121 targets the V3 glycan site. The combination leverages their complementary neutralization profiles to provide broader and more potent coverage against diverse global HIV-1 strains. Clinical studies have shown that this dual bNAb regimen is generally safe and well tolerated, with pharmacokinetic data indicating median elimination half-lives of approximately 25 days for PGDM1400 and 32 days for PGT121. The combination has been evaluated in phase 1 clinical trials as both a dual therapy (PGDM1400 + PGT121) and as part of triple combinations with other bNAbs[4][5]. The primary indication under investigation is HIV-1 infection.
