Overview

Phesgo is a fixed-dose combination of pertuzumab, trastuzumab, and hyaluronidase-zzxf administered subcutaneously. Pertuzumab and trastuzumab are monoclonal antibodies that target the human epidermal growth factor receptor 2 (HER2), blocking different sites on the HER2 receptor to inhibit cancer cell growth and survival. Hyaluronidase-zzxf increases tissue permeability to facilitate subcutaneous absorption. Exemestane is an oral steroidal aromatase inhibitor that suppresses estrogen synthesis by irreversibly binding to the aromatase enzyme, reducing estrogen levels in postmenopausal women. The combination of Phesgo with exemestane is used as a first-line treatment for metastatic HER2-positive breast cancer, particularly in hormone receptor-positive cases where endocrine therapy is indicated[2][5]. This regimen leverages dual HER2 blockade with hormonal suppression for enhanced antitumor activity.
