phosphate-buffered saline + Incomplete Freund's adjuvant is an experimental control formulation used in immunological and oncological research, specifically within preclinical vaccine studies. It consists of an aqueous phosphate-buffered saline (PBS) phase emulsified with Incomplete Freund's adjuvant (IFA), which is a mixture of paraffin oil and an emulsifying agent (typically mannide monooleate). Unlike Complete Freund's Adjuvant (CFA), IFA does not contain heat-killed mycobacteria, making it less inflammatory while still providing a 'depot' effect. This effect allows for the slow release of substances from the injection site, enhancing the recruitment and activation of antigen-presenting cells. In the context of small cell lung cancer (SCLC) research, this combination is utilized as a negative control to establish baseline survival and immune response metrics against which experimental neo-antigen vaccines, such as those targeting isoaspartylated ELAVL4, are measured.
