phosphoprotein 65 messenger RNA-lipid particles + tumor messenger RNA-lipid particles is an experimental personalized mRNA vaccine combination currently under investigation for the treatment of high-grade gliomas, including glioblastoma multiforme (GBM). The therapy utilizes lipid nanoparticles (LNPs) to deliver two distinct sets of mRNA: one encoding the immunodominant Cytomegalovirus (CMV) antigen phosphoprotein 65 (pp65), and another derived from the patient's own tumor (total tumor RNA). The rationale for this approach is based on the observation that CMV antigens like pp65 are frequently expressed in GBM tissues but not in the surrounding normal brain, providing a specific target for immunotherapy. By combining this with autologous tumor RNA, the vaccine aims to trigger a broad and potent T-cell mediated immune response against both viral and patient-specific tumor-associated antigens, overcoming the immunosuppressive microenvironment of the central nervous system. This approach is primarily associated with clinical research conducted at the University of Florida.
