PHY906 + capecitabine is a combination therapy consisting of capecitabine, an oral prodrug of 5-fluorouracil (5-FU) and a widely used chemotherapeutic agent, and PHY906, a standardized pharmaceutical-grade formulation derived from four traditional Chinese medicinal herbs. Capecitabine acts as an antimetabolite that inhibits DNA synthesis in rapidly dividing cancer cells. PHY906 is designed to enhance the therapeutic index of chemotherapeutic agents by both potentiating their anti-tumor activity and reducing gastrointestinal toxicity. Preclinical studies have shown that PHY906 can increase the efficacy of various chemotherapy drugs—including capecitabine—while also alleviating side effects such as diarrhea and mucosal injury. Clinical trials in patients with advanced hepatocellular carcinoma (HCC) have demonstrated that this combination may improve disease control rates and overall survival compared to chemotherapy alone, particularly in certain patient subgroups such as those with hepatitis B virus infection or lower alpha-fetoprotein levels[1][2][3]. The mechanism by which PHY906 exerts its effects includes anti-inflammatory actions (e.g., inhibition of nuclear factor kappaB, cyclooxygenase-2), promotion of intestinal epithelial regeneration via Wnt signaling pathways, and modulation of immune responses[6][9].
