A combination therapy consisting of a phosphoinositide 3-kinase (PI3K) inhibitor and a selective estrogen receptor degrader (SERD). This regimen is primarily under investigation for the treatment of hormone receptor-positive, HER2-negative advanced or metastatic breast cancer, particularly in patients with PIK3CA mutations who have progressed on prior endocrine therapies. The PI3K inhibitor component targets the PI3K/AKT/mTOR signaling pathway, which is frequently activated in breast cancer and drives cell proliferation and survival. The SERD component binds to and degrades the estrogen receptor, blocking estrogen-driven tumor growth. This combination aims to overcome resistance mechanisms that limit the efficacy of single-agent therapies by simultaneously inhibiting two critical pathways involved in tumor progression[1][2][5][6].
