Overview

Piamprilimumab + anlotinib is an investigational combination therapy being evaluated for the treatment of advanced solid tumors. Piamprilimumab (SCT-I10A) is a humanized IgG4 monoclonal antibody that acts as a checkpoint inhibitor by binding to the programmed cell death protein 1 (PD-1) receptor, thereby preventing its interaction with ligands PD-L1 and PD-L2 and restoring anti-tumor T-cell activity. Anlotinib is an orally administered, multi-targeted small molecule tyrosine kinase inhibitor that suppresses tumor angiogenesis and growth by inhibiting vascular endothelial growth factor receptors (VEGFR1, VEGFR2, VEGFR3), fibroblast growth factor receptors (FGFR1-4), platelet-derived growth factor receptors (PDGFRα, PDGFRβ), and c-Kit. This combination is primarily developed and studied in China, with Shanxi Bethune Hospital serving as a prominent clinical trial site for Phase II studies in indications such as esophageal squamous cell carcinoma and gastric cancer. The rationale for the combination is to leverage the synergistic effects of immune checkpoint blockade and anti-angiogenic therapy to overcome tumor-induced immunosuppression and improve clinical outcomes in patients with refractory solid tumors.
