**Pimasertib + temsirolimus** is an experimental combination therapy investigated for the treatment of advanced solid tumors. The combination employs dual pathway inhibition: pimasertib is a selective inhibitor of MEK 1/2 (mitogen-activated protein kinase kinases 1 and 2), suppressing the MAPK/ERK signaling pathway, while temsirolimus is an mTOR (mechanistic target of rapamycin) inhibitor, suppressing the PI3K/AKT/mTOR signaling pathway. Preclinical rationale supported the potential for synergistic antitumor effects by targeting two critical survival and proliferation pathways frequently co-activated in cancers. However, a phase I clinical trial in patients with advanced solid tumors found that this combination resulted in frequent dose-limiting toxicities (primarily stomatitis and thrombocytopenia) and overlapping toxicities that precluded definition of a recommended phase II dose (RP2D). No further development was pursued for this combination due to safety and tolerability concerns[1][2][3].
