PIN + sintilimab is an investigational combination therapy consisting of a Pyroptosis-inducible Newcastle Disease Oncolytic Virus (PIN) and the PD-1 checkpoint inhibitor sintilimab. PIN is an oncolytic virus engineered to selectively infect and replicate within tumor cells, triggering pyroptosis—a highly inflammatory form of programmed cell death. This process releases tumor-associated antigens and pro-inflammatory cytokines, effectively converting the tumor microenvironment from "cold" to "immunologically hot." Sintilimab is a humanized monoclonal antibody that blocks the interaction between the Programmed Cell Death Protein 1 (PD-1) receptor on T cells and its ligands (PD-L1/PD-L2), thereby preventing immune evasion. The combination is specifically being evaluated by the Chinese PLA General Hospital for the treatment of advanced proficient mismatch repair/microsatellite stable (pMMR/MSS) colorectal cancer with hepatic metastases, a population that typically shows poor response to single-agent checkpoint inhibition.
