Overview

Pioglitazone + exenatide is a fixed-dose combination of two antidiabetic agents used for the treatment of type 2 diabetes mellitus. Pioglitazone is a thiazolidinedione that acts as an agonist at peroxisome proliferator-activated receptor gamma (PPARγ), improving insulin sensitivity in adipose tissue, muscle, and the liver. Exenatide is a glucagon-like peptide-1 (GLP-1) receptor agonist that enhances glucose-dependent insulin secretion, suppresses inappropriate glucagon secretion, slows gastric emptying, reduces appetite, and promotes weight loss. The combination targets both insulin resistance and beta-cell dysfunction—the core defects in type 2 diabetes—resulting in improved glycemic control with a low risk of hypoglycemia. Clinical studies have shown this combination to be effective and safe for patients with poorly controlled or newly diagnosed type 2 diabetes[3][5][8].
