pioglitazone + rosiglitazone

Pioglitazone + rosiglitazone is a hypothetical combination of two thiazolidinedione-class oral antidiabetic agents, pioglitazone and rosiglitazone. Both drugs act as insulin sensitizers by activating peroxisome proliferator-activated receptor gamma (PPARγ), thereby improving glucose uptake in adipose tissue, muscle, and the liver. Pioglitazone also has some activity at PPARα, which may contribute to its effects on lipid metabolism[3][7]. These agents are primarily indicated for the management of type 2 diabetes mellitus as adjuncts to diet and exercise[1][2][5]. While both drugs share similar mechanisms of action, they differ slightly in their receptor selectivity and safety profiles; notably, concerns have been raised about increased cardiovascular risk with rosiglitazone compared to pioglitazone[6][10]. There is no evidence that a fixed-dose or co-formulated product containing both pioglitazone and rosiglitazone exists or is approved for use.
