piperaquine + pyronaridine + artesunate is an experimental triple antimalarial combination therapy being evaluated by the University of Oxford in collaboration with the Global Health Innovative Technology Fund (GHIT Fund). This combination therapy consists of piperaquine, a long-acting bisquinoline antimalarial, and Pyramax, which is a fixed-dose combination of pyronaridine tetraphosphate and artesunate. The triple combination (TACT) is designed to provide a robust treatment regimen that can overcome multidrug-resistant Plasmodium falciparum malaria. Piperaquine and pyronaridine both act by inhibiting the detoxification of heme within the parasite's food vacuole, while artesunate provides rapid parasite clearance through the generation of reactive free radicals. A Phase 1, open-label, randomized crossover study (NCT06114108) is currently being conducted to evaluate the pharmacokinetic interactions and safety of these three orally administered agents in healthy adult participants.
