Overview
Pirotinib (also known as pirotinib hydrochloride or XZP-3287) is an orally bioavailable, small molecule irreversible pan-epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor. Developed by Sihuan Pharmaceutical Holdings Group through its subsidiary, Shandong Xuanzhu Pharmaceutical Technology, pirotinib acts as an antagonist against EGFR (erbB1), HER2 (erbB2), and HER4 (erbB4). By covalently binding to these receptors, it blocks the downstream signaling pathways that drive tumor cell proliferation and survival. Pirotinib is primarily investigated for the treatment of advanced non-small cell lung cancer (NSCLC), particularly in patients whose tumors harbor specific genomic variants. It has been evaluated in the Phase II TRUMP umbrella trial, sponsored by the Guangdong Association of Clinical Trials, which utilizes next-generation sequencing (NGS) to match patients with targeted therapies.
