Overview

K-924 LD is a low-dose fixed-dose combination (FDC) tablet containing pitavastatin calcium (2 mg) and ezetimibe (10 mg), developed by Kowa Company, Ltd. for the treatment of hypercholesterolemia. Pitavastatin is a potent HMG-CoA reductase inhibitor (statin) that reduces endogenous cholesterol production in the liver by inhibiting the rate-limiting enzyme in the biosynthetic pathway. Ezetimibe is a selective cholesterol absorption inhibitor that targets the Niemann-Pick C1-Like 1 (NPC1L1) transporter in the small intestine, thereby reducing the uptake of dietary and biliary cholesterol. The combination is designed to provide superior LDL-C lowering efficacy compared to pitavastatin monotherapy by simultaneously addressing two distinct pathways of cholesterol regulation. K-924 LD was evaluated in Phase III clinical trials in Japan and subsequently approved under the brand name Livalozet.
