Overview

PL9654 and PL9655 are proprietary small molecule melanocortin receptor (MCR) agonists being developed by Palatin Technologies in collaboration with Boehringer Ingelheim. These compounds are designed to treat retinal diseases, specifically diabetic retinopathy and diabetic macular edema, by targeting the melanocortin receptor system. Their mechanism of action involves activating natural resolution pathways to reduce inflammation, stabilize the blood-retinal barrier, decrease vascular endothelial growth factor (VEGF) signaling, and provide neuroprotection to retinal ganglion cells. The program is currently in the preclinical stage of development following a strategic partnership announced in August 2025.
