Overview

This is a combination of two agents: placebo and norethisterone. Placebo refers to an inert substance with no pharmacological activity, commonly used as a control in clinical trials. Norethisterone (also known as norethindrone) is a synthetic progestin, part of the progestogen class, that mimics the effects of natural progesterone. Norethisterone acts primarily by binding to progesterone receptors in target tissues such as the reproductive tract, breast, pituitary gland, hypothalamus, skeletal tissue, and central nervous system. Its main mechanisms include suppression of ovulation via negative feedback on gonadotropin release (FSH and LH), alteration of cervical mucus to impede sperm transport, and induction of endometrial changes that prevent implantation[1][2][5]. It is used for contraception (as a "mini pill"), treatment of menstrual disorders (such as heavy or irregular periods), endometriosis management, premenstrual syndrome relief, hormone replacement therapy in menopause (with estrogen), and sometimes for delaying menstruation or treating breast cancer[1][2][3][4][5].
