Drug intelligence / Profile preview
The RTS,S malaria vaccine (marketed as Mosquirix) is a recombinant protein-based, virus-like particle (VLP) vaccine developed by GSK in collaboration with the Walter Reed Army Institute of Research, PATH, and Bharat Biotech. It is the first malaria vaccine to receive regulatory approval and WHO recommendation for the prevention of Plasmodium falciparum malaria in children living in moderate-to-high transmission regions. The vaccine consists of a fusion protein containing a portion of the P. falciparum circumsporozoite protein (CSP) fused to the hepatitis B surface antigen (HBsAg), co-expressed with free HBsAg in Saccharomyces cerevisiae to form VLPs. It is formulated with the AS01 adjuvant system (containing QS-21, MPL, and liposomes) to enhance immunogenicity. RTS,S primarily works by inducing high titers of anti-CSP antibodies and CD4+ T-cell responses that target the pre-erythrocytic sporozoite stage, preventing the parasite from infecting hepatocytes and establishing a blood-stage infection.
