Overview

RTS,S/AS01E (marketed as Mosquirix) is a recombinant protein-based malaria vaccine developed by GSK in collaboration with the Walter Reed Army Institute of Research and the PATH Malaria Vaccine Initiative. It is the first malaria vaccine recommended by the World Health Organization (WHO) for broad use in children living in regions with moderate to high malaria transmission. The vaccine consists of a fusion protein containing portions of the Plasmodium falciparum circumsporozoite protein (CSP) fused to the hepatitis B surface antigen (HBsAg), which self-assembles into virus-like particles. It is formulated with GSK's proprietary AS01E adjuvant system, which contains monophosphoryl lipid A (MPL) and QS-21 in a liposomal formulation, to enhance the immune response. RTS,S/AS01E targets the pre-erythrocytic stage of the malaria parasite, inducing antibodies and T-cell responses that block sporozoites from infecting hepatocytes in the liver.
