Platinum + etoposide + anlotinib is a combination regimen used primarily as first-line therapy for extensive-stage small-cell lung cancer (ES-SCLC). The regimen consists of a platinum-based chemotherapy agent (cisplatin or carboplatin), the topoisomerase II inhibitor etoposide, and anlotinib, which is a novel oral multi-targeted tyrosine kinase inhibitor. Anlotinib inhibits angiogenesis by targeting multiple receptors including vascular endothelial growth factor receptor 2 (VEGFR2), vascular endothelial growth factor receptor 3 (VEGFR3), fibroblast growth factor receptor 1 (FGFR1), fibroblast growth factor receptor 2 (FGFR2), fibroblast growth factor receptor 3 (FGFR3), fibroblast growth factor receptor 4 (FGFR4), platelet-derived growth factor receptor alpha (PDGFRα), platelet-derived growth factor receptor beta (PDGFRβ), c-kit, and Ret. This combination has demonstrated improved progression-free survival and overall response rates compared to traditional chemotherapy alone in clinical studies for ES-SCLC. Anlotinib’s antiangiogenic activity provides synergistic effects with cytotoxic agents, while its toxicity profile allows for tolerable combination use[1][2][6][3].
