PLN-101095 + pembrolizumab is an investigational combination therapy for adult patients with advanced or metastatic solid tumors who have progressed after prior treatment with pembrolizumab. PLN-101095 is a novel small-molecule inhibitor targeting integrins αvβ1 and αvβ8, designed to modulate the tumor microenvironment and potentially enhance immune recognition. Pembrolizumab is a well-established monoclonal antibody against programmed cell death protein 1 (PD-1), used as an immune checkpoint inhibitor to promote antitumor immune responses. The rationale for this combination is that PLN-101095 may overcome resistance to pembrolizumab by altering the tumor stroma and improving immune infiltration, and the regimen is currently being studied in a dose-escalation Phase 1 clinical trial for patients with solid tumors refractory or relapsed to pembrolizumab therapy[1][2][3][4].
