PLX2853 + carboplatin is a combination therapy of **PLX2853**, an orally active small molecule **BET (bromodomain and extra-terminal domain) inhibitor**, with **carboplatin**, a platinum-based chemotherapy agent. PLX2853 works by selectively inhibiting BET protein interactions, interfering with epigenetic regulation that drives cancer cell growth, particularly in tumors with *ARID1A* mutations, which are common in certain gynecologic malignancies. Carboplatin induces DNA crosslinking and apoptosis in rapidly dividing cells. This combination is primarily being studied in women with platinum-resistant epithelial ovarian cancer and in gynecological cancers harboring ARID1A mutations, with the hypothesis that BET inhibition may reverse platinum resistance or act synergistically with carboplatin[1][2][4][5].
