PM1021 + PM8001 is an investigational combination immunotherapy currently in early-stage clinical trials for advanced solid tumors. PM1021 is a recombinant human immunoglobulin G1 (IgG1) monoclonal antibody targeting TIGIT (T-cell immunoreceptor with Ig and ITIM domains), an immune checkpoint inhibitor involved in tumor immune evasion[4][3][1]. PM8001 is a bifunctional protein composed of the extracellular domain of the TGF-β receptor II (TGF-β "trap") fused to a humanized anti-PD-L1 IgG1 single-domain antibody, simultaneously inhibiting both the PD-L1/PD-1 pathway and neutralizing TGF-β in the tumor microenvironment[2][3][6][5]. This dual blockade aims to enhance anti-tumor immune responses by counteracting multiple mechanisms of immune suppression. The combination is developed by Biotheus and is undergoing phase I evaluation as a treatment for patients with advanced metastatic cancers that have failed standard therapies[1][3][5][6].
