Drug intelligence / Profile preview
PMN440 (formerly PMN400) is an investigational therapeutic vaccine being developed by ProMIS Neurosciences for the treatment of synucleinopathies, including Parkinson's disease, multiple system atrophy (MSA), and dementia with Lewy bodies (DLB). Designed using ProMIS's proprietary EpiSelect computational platform, the vaccine consists of computationally-derived conformational B cell epitopes from the EKTKEQ region (amino acids 57-62) of misfolded pathogenic alpha-synuclein. These peptide epitopes are conjugated to keyhole limpet hemocyanin (KLH) and formulated with the saponin-based adjuvant QS-21. PMN440 is engineered to selectively elicit high-affinity antibodies against toxic, misfolded alpha-synuclein oligomers and soluble seeding fibrils, while sparing physiological monomers and non-toxic insoluble fibril deposits, thereby preserving normal alpha-synuclein function and avoiding target distraction.
