Overview
Pneumococcal 21-valent conjugate vaccine (V116) is a polysaccharide conjugate vaccine developed by Merck (MSD) for the prevention of invasive disease and pneumonia caused by Streptococcus pneumoniae. It is specifically designed to address the epidemiology of pneumococcal disease in adults, covering 21 serotypes that account for approximately 84% of invasive pneumococcal disease in individuals aged 65 and older. The vaccine includes eight unique serotypes (15A, 15C, 16F, 23A, 23B, 24F, 31, and 35B) not found in other licensed pneumococcal vaccines. Each polysaccharide is conjugated to the CRM197 carrier protein to elicit a T-cell dependent immune response, which provides superior immunogenicity and immunological memory compared to pure polysaccharide vaccines. The Leids Universitair Medisch Centrum (LUMC) has been a key collaborator in evaluating the vaccine's efficacy using controlled human infection models.
