Overview

pneumococcal polysaccharide serotype 3 conjugate (CRM197) adsorbed on aluminium phosphate is a monovalent pneumococcal conjugate vaccine (PCV) candidate developed by the Rijksinstituut voor Volksgezondheid en Milieu (RIVM). It is specifically designed to target Streptococcus pneumoniae serotype 3, a pathogen known for its high virulence and the relative difficulty in achieving high vaccine efficacy compared to other serotypes in multivalent formulations. The vaccine utilizes the CRM197 carrier protein, a non-toxic mutant of the diphtheria toxin, to conjugate the serotype 3 capsular polysaccharide. This conjugation converts the T-cell independent polysaccharide antigen into a T-cell dependent antigen, thereby eliciting a more robust immune response and providing immunological memory. The formulation is adsorbed on aluminium phosphate, which serves as an adjuvant to further enhance immunogenicity. This candidate was developed as part of RIVM's broader initiative to create affordable pneumococcal vaccines and facilitate technology transfer to manufacturers in low- and middle-income countries.
