Overview

pneumococcal polysaccharide serotype 7F conjugate (CRM197) adsorbed on aluminium phosphate is a monovalent vaccine component developed by the Rijksinstituut voor Volksgezondheid en Milieu (RIVM) in the Netherlands. It is designed to provide active immunity against *Streptococcus pneumoniae* serotype 7F, a common cause of invasive pneumococcal diseases such as pneumonia, meningitis, and bacteremia. The vaccine utilizes a conjugate technology where the purified capsular polysaccharide of serotype 7F is covalently linked to CRM197, a non-toxic recombinant mutant of the diphtheria toxin. This conjugation is critical for converting the polysaccharide from a T-cell independent to a T-cell dependent antigen, which allows for the induction of immunological memory and a robust immune response in infants and young children. The conjugate is adsorbed onto an aluminium phosphate adjuvant to further enhance its immunogenicity. This specific conjugate technology was developed as part of a global health initiative to create affordable pneumococcal conjugate vaccines (PCVs) and has been licensed to manufacturers like the Serum Institute of India for inclusion in multivalent vaccines such as Pneumosil.
