Overview

It is designed for the treatment of pain conditions involving both nociceptive and neuropathic components, such as chronic low-back pain and severe to moderate pain with inflammatory elements. The combination aims to leverage the analgesic and anti-inflammatory effects of polmacoxib and the neuropathic pain control of pregabalin, providing enhanced efficacy and improved dosing convenience compared to monotherapy with either agent. Polmacoxib inhibits prostaglandin synthesis by selectively targeting COX-2, thereby reducing pain and inflammation, while pregabalin decreases neuronal excitability and neurotransmitter release by modulating calcium channel activity and inhibiting calcium influx at presynaptic nerve terminals[2][3][4]. No known pharmacological interaction between the two agents in the fixed-dose combined form based on pharmaceutical formulation studies[3].
