Poly(2-ethyl-2-oxazoline) (PEtOx) is a biocompatible, water-soluble synthetic polymer belonging to the poly(2-oxazoline) family. It is widely investigated as a functional alternative to poly(ethylene glycol) (PEG) in pharmaceutical and biomedical applications due to its "stealth" properties, low immunogenicity, and high chemical stability. PEtOx is utilized as a versatile drug delivery platform, including as a component in lipid nanoparticles (LNPs), long-acting injectable microspheres, and amorphous solid dispersions to enhance the solubility and bioavailability of poorly water-soluble drugs. In veterinary medicine, PEtOx-conjugated porcine serum albumin (POx-PSA) has been developed as a therapeutic agent to maintain circulating blood volume in cases of hemorrhagic shock. Additionally, the polymer is used in the fabrication of medical devices, such as electrospun grafts for tympanic membrane repair, and as a safety-assessed additive in food contact materials.
