**poly-ICLC + atezolizumab** is a combination therapy consisting of poly-ICLC (polyinosinic-polycytidylic acid stabilized with poly-L-lysine and carboxymethylcellulose), an immunostimulatory TLR3 agonist, and atezolizumab, a monoclonal antibody immune checkpoint inhibitor targeting PD-L1. Poly-ICLC functions as an in situ vaccine adjuvant by activating innate immunity via Toll-like receptor 3 (TLR3), promoting dendritic cell maturation, and enhancing antigen presentation, thereby priming adaptive immune responses. Atezolizumab blocks PD-L1, preventing the inhibition of cytotoxic T lymphocytes and enabling improved antitumor immune activity. When administered in combination, poly-ICLC acts as an immunogenic stimulant, and atezolizumab augments checkpoint blockade, yielding improved systemic antitumor immune responses[2][3]. Clinical investigation focuses on their use in unresectable hepatocellular carcinoma and other advanced cancers where immune modulation is beneficial[3].
