Overview

**poly-ICLC + cemiplimab** is an experimental combination therapy consisting of poly-ICLC (Polyinosinic-polycytidylic acid stabilized with poly-L-lysine and carboxymethylcellulose, known commercially as Hiltonol), a synthetic double-stranded RNA immunostimulant, and cemiplimab (Libtayo), a human IgG4 monoclonal antibody targeting PD-1. Poly-ICLC acts as a toll-like receptor 3 (TLR3) agonist, stimulating both innate and adaptive immune responses, and is explored for its vaccine adjuvant and direct anti-tumor immunostimulant effects. Cemiplimab blocks the programmed cell death 1 (PD-1) receptor, releasing T-cell inhibition and enabling anti-tumor immune response. The combination is under investigation for treating accessible solid tumors, including cases where anti-PD-1/PD-L1 checkpoint blockade is combined with poly-ICLC to potentially enhance anti-tumor activity[2][5].
