Overview

TPC is a combination induction chemotherapy regimen consisting of a polymeric micellar formulation of paclitaxel, cisplatin, and capecitabine. Developed and studied by researchers at Sun Yat-sen University, this regimen is specifically investigated for high-risk locoregionally advanced nasopharyngeal carcinoma (NPC). The polymeric micellar paclitaxel component is a Cremophor EL-free nanoparticle formulation designed to enhance tumor penetration via the enhanced permeability and retention (EPR) effect while minimizing the hypersensitivity reactions typically associated with standard paclitaxel. Within the regimen, paclitaxel functions by stabilizing microtubules and inhibiting mitosis, cisplatin acts as a DNA-alkylating-like agent to induce apoptosis, and capecitabine serves as an oral fluoropyrimidine carbamate that is enzymatically converted to 5-fluorouracil, which inhibits thymidylate synthase and interferes with DNA synthesis.
