This is a combination drug consisting of three active ingredients—polymyxin B, gentamicin, and amphotericin B. Each component has a distinct mechanism of action: - **Polymyxin B** is a polypeptide antibiotic that disrupts the cell membrane integrity of susceptible Gram-negative bacteria by binding to lipopolysaccharides and phospholipids in the outer cell membrane, leading to increased permeability and cell death[1]. - **Gentamicin** is an aminoglycoside antibiotic that binds irreversibly to the 30S subunit of bacterial ribosomes, inhibiting protein synthesis and causing misreading of mRNA in susceptible bacteria[3][5]. - **Amphotericin B** is an antifungal agent that binds to ergosterol in fungal cell membranes, forming pores that increase membrane permeability and result in leakage of intracellular components and fungal cell death[2]. This combination may be used for topical or local treatment where mixed infections (bacterial including Gram-negative organisms, as well as fungal) are suspected or confirmed.
