Overview
PolyPEPI1018 + atezolizumab is an investigational combination regimen consisting of PolyPEPI1018, a multi-peptide cancer vaccine, and atezolizumab, a humanized anti–PD-L1 monoclonal antibody (marketed as Tecentriq). PolyPEPI1018 is composed of 12 immunogenic peptides derived from seven tumor-associated antigens designed to induce polyclonal T-cell responses against tumor cells. Atezolizumab blocks the interaction between PD-L1 and its receptors (PD-1 and B7.1), releasing the inhibition of the immune response and promoting immune-mediated tumor cell killing. This combination aims to convert immunologically cold, microsatellite-stable (MSS) metastatic colorectal cancer (mCRC) tumors into immunologically active ("hot") ones, improving immune infiltration and potentially antitumor efficacy in a setting where checkpoint inhibitor monotherapy typically shows poor benefit.
