PolyPhetamine is a polymer-drug conjugate being developed by ITL Pharma for the treatment of Attention Deficit Hyperactivity Disorder (ADHD). The drug utilizes a proprietary delivery technology where amphetamine is chemically bonded to a polymer backbone, creating a prodrug that requires enzymatic or physiological cleavage for activation. This design is intended to provide a smooth, extended-release pharmacokinetic profile and reduce the potential for drug abuse, as the stimulant cannot be easily extracted or rapidly released through non-oral routes. Once released, the amphetamine moiety acts by increasing synaptic concentrations of dopamine and norepinephrine through the stimulation of trace amine-associated receptor 1 (TAAR1) and the inhibition of the dopamine and norepinephrine transporters.
