Overview

This combination consists of **pomalidomide**, an immunomodulatory agent, with **cisplatin** and **etoposide**, both cytotoxic chemotherapeutic agents. As reported in phase I and II studies, this regimen was evaluated in treatment-naive patients with extensive-stage small-cell lung cancer (ES SCLC) and is also used in relapsed/refractory multiple myeloma as part of salvage regimens. Pomalidomide modulates the immune system and has antiangiogenic and anti-neoplastic properties, acting by enhancing T-cell and natural killer (NK) cell activation and inhibiting pro-inflammatory cytokines. Cisplatin forms DNA crosslinks resulting in apoptosis, while etoposide inhibits topoisomerase II, leading to DNA strand breaks and cell death[1][3][4]. The combination is experimental; its primary clinical investigation has been in SCLC and heavily pre-treated myeloma or plasma cell leukemia.
