Overview

**pomalidomide + marizomib + dexamethasone** is an investigational combination therapy evaluated for the treatment of relapsed or refractory multiple myeloma. Pomalidomide is a third-generation immunomodulatory drug approved for multiple myeloma, acting primarily by modulating immune responses and affecting tumor microenvironment. Marizomib is an intravenous, irreversible pan-proteasome inhibitor developed to disrupt protein degradation mechanisms essential for multiple myeloma cell survival. Dexamethasone is a synthetic glucocorticoid corticosteroid used to suppress inflammation and immune responses, and to enhance the effectiveness of other anti-myeloma agents. This three-drug regimen is designed to exploit multiple anti-myeloma mechanisms by targeting the proteasome, modulating immunity, and suppressing inflammation. The combination has demonstrated promising activity in heavily pretreated, relapsed/refractory multiple myeloma patients in early-phase clinical studies[1][2][5][6].
