Overview

Ponatinib + blinatumomab is a combination regimen of two distinct pharmaceuticals used primarily for the treatment of Philadelphia chromosome-positive (Ph+) acute lymphoblastic leukemia (ALL). Ponatinib is an oral small molecule tyrosine kinase inhibitor that targets BCR-ABL1 and other kinases, while blinatumomab is a bispecific T-cell engager antibody construct that directs cytotoxic T cells to CD19-expressing B cells. This chemotherapy-free combination has demonstrated high rates of complete molecular response and minimal residual disease negativity in both newly diagnosed and relapsed/refractory Ph+ ALL patients. The regimen may reduce the need for allogeneic stem cell transplantation and mitigate toxicities associated with traditional chemotherapy[1][3][4][5][6].
