PORT-6 + Keytruda is a combination therapy being developed by Portage Biotech in collaboration with Merck for the treatment of advanced solid tumors. The regimen combines PORT-6 (formerly TT-10), a potent and selective small molecule antagonist of the adenosine 2A receptor (A2AR), with Keytruda (pembrolizumab), an anti-PD-1 monoclonal antibody. This combination is designed to target two key mechanisms of immune evasion in the tumor microenvironment: PORT-6 blocks the immunosuppressive signaling of adenosine, which is often elevated in tumors, while Keytruda inhibits the PD-1/PD-L1 checkpoint pathway. By simultaneously relieving adenosine-driven suppression and restoring T-cell activity, the therapy aims to enhance the immune system's ability to recognize and destroy cancer cells. It is currently being evaluated in the ADPORT-601 Phase 1a/1b clinical trial, focusing on patients with A2AR-high solid tumors.
